چکیده

The aim of the study was to characterise the free coagulase-negative Staphylococcus sp. strains isolated from normal (9.4%) and inflamed quarter milk samples of dairy cows in one herd localised in southern Poland. The isolates were examined phenotypically by cultural features, tube coagulase test, clumping factor (CF), and commercial agglutination test. Total number of 427 strains were cultivated on different media: Congo red agar, DNase medium, mannitol salt agar, and crystal violet agar. About 25% of the strains were examined by the disc diffusion method for their sensitivity to chosen antibiotics. All of the analysed strains showed α, β, α+β, or lh type of haemolysis on plate with 5% sheep blood agar, and had positive reaction in CF and agglutination tests, but were negative in tube coagulase test. These strains showed DNase activity and ability to mannitol fermentation; however, none of the strains produced slime. There were variations in growth on crystal violet agar: examined strains belonged to three types of growth (A, E, and C). The strains were highly susceptible in vitro to oxacillin (99.2%), cefoperazone (95.2%), and amoxicillin with clavulanic acid (93.6%), and highly resistant to neomycin (27.2%), tetracycline (21.6%), and streptomycin (20.8%). In conclusion, it can be stated that intramammary infections in cows can be caused by atypical free coagulase-negative strains of Staphylococcus aureus.

Molecular typing of Staphylococcus aureus based on PCR-RFLP of coa gene and RAPD analysis.

The aim of this study was molecular identification of S. aureus strains isolated from mastitic milk samples and establishing the genetic relationship between strains isolated from cows belonging to the same herd. In all 43 isolated strains the gap gene (930 bp) was amplified, which enabled their affiliation to the Staphylococcus genus to be established. PCR-RFLP with AluI endonuclease of the ga...

Enterotoxin gene profiles among Staphylococcus aureus isolated from raw milk

Milk is considered a nutritious food because it contains several important nutrients including proteins and vitamins. Conversely, it can be a vehicle for several pathogenic bacteria such as Staphylococcus aureus. This study aimed to analyze the frequency of genes encoding the nine Staphylococcal enterotoxins (SEs) and enterotoxin gene profiles in S. aureus isolates derived from raw bovine milk....

Antibiotic Susceptibility and Plasmid Profiles of Staphylococcus aureus Strains Isolated from Bovine Subclinical Mastitis in Tabriz, Iran

ABSTRACT        Background and Objectives: Mastitis caused by Staphylococcus aureus is a widely distributed disease in cattle, goats and sheep. The infection is often subclinical in cattle, leading to reduced milk production and quality, but acute catarrhal or even gangrenous inflammation may also occur. The aim of this study was to investigate resistance of S. au...

Phenotypical and Genotypical Assessment Techniques for Identification of Some Contagious Mastitis Pathogens

Mastitis is one of the most economic disease affecting dairy cows worldwide. Its classic diagnosis using bacterial culture and biochemical findings is a difficult and prolonged method. In this research, using of matrix-assisted laser desorption/ionization time-offlight mass spectrometry (MALDI-TOF MS) permitted identification of different microorganisms with high accuracy and rapidity (only 24 ...

Atypical Staphylococcus Aureus as an Aetiological Agent of Mastitis in Cows

The aim of the study was to characterise the 493 tube-coagulase-negative Staphylococcus sp. strains isolated from the udder quarters of milking dairy cows in one herd with a high bulk milk somatic cell count (SCC). The isolates were examined phenotypically by cultural features, Gram stain, catalase, coagulase, and API biochemical tests, and genotypically by PCR for chosen genes. About 10% of th...
